Saposin C (SAP-C) is a small, heat-stable lysosomal glycoprotein derived from the prosaposin precursor protein via site-specific proteolytic cleavage. Among the four prosaposin-derived domains, Saposin C shares high structural homology with Saposin A and acts as an indispensable biological cofactor required for the lysosomal activation of β-glucocerebrosidase (GBA)—the enzyme deficient in Gaucher disease. Additionally, Saposin C plays a critical role in cellular immunology by facilitating the presentation of lipid antigens through CD1b molecules to human T cells. It can be assembled with dioleoylphosphatidylserine to form SapC-DOPS, a highly selective, membrane-targeted nanocarrier platform used to target and induce apoptosis in various cancer models, including glioblastoma multiforme.
